Mice(male Kunming mice;500.00 mg kg−1 day−1 D-gal;hypodermic injection;8 weeks)

The mice had free access to food and water during the week-long of adaptive feeding. After one week of acclimatization, the mice were randomly divided into 5 groups (Blank,Model, WO, PSO, CSO). The first week of the experiment, apart from the blank group (treated with saline), all the mice in the 4 other groups received 500.00 mg kg−1 day−1 D-gal by hypodermic injection for 8 weeks. The first two weeks, all groups were supplied by maintenance fodder.

Mice(male Kunming mice;300 mg/kg ;D-gal;intraperitoneally injected;42 days)

The experimental animals used in this experiment were Kunming mice (n = 36, male, 8-week-old, 25 ± 4 g), provided by the Laboratory Animal Center of Dali University. The mice were kept in a relatively constant environment (temperature 25 ± 1 ◦C, humidity 60 % ± 5 %) with alternating light and dark cycles for 12 h. After adaptation, the patients were randomly divided into control group (saline, n = 12), D-gal group (300 mg/kg D-gal, n = 12) and GAS group (300 mg/kg D-gal+78 mg/kg gastrodin injection, n = 12). D-gal group and GAS group were intraperitoneally injected with D-gal once a day for 42 days to establish the aging mouse model, and mice in control group were intraperitoneally injected with the same amount of saline every day.

Mice(ICR;6 weeks old;1 % D-galactose;subcutaneous injection;28 days)

A total of 60 mice (ICR, SPF grade, male, 6 weeks old, weight 28.0 ±1.5 g) were randomly and equally divided into control, D-galactose(Solarbio, batch No. 1013I056), 0.2 g/kg metformin, 3.6 g/kg ginseng water decoction, 1.8 g/kg GWD , and 0.9 g/kg GWD (half equivalent dose) groups. The dosage was calculated by multiplying 12.3 according to the equivalent dose coefficient of human and mouse body surface area in “Pharmacological Experimental Methodology.” The quality control of GWD was the same as previous study (Wu et al., 2020). According to the dosing volume of 10 mL/kg,mice were injected with 1 % D-galactose aqueous solution subcutaneously on the back of the neck to establish an aging model for 28 days.

Mice(male Kunming mice;ip;52 days)

The AD model mice were replicated by injection of D-gal intraperitoneally and fed with high-fat diet for 56 consecutive days. The AD model mice were orally administered with high dosed BYHWD at 58.58 g/kg/d calculated with herbs (BHD-H
group), middle-dosed BYHWD at 29.29 g/kg/d (BHD-M group), low-dosed BYHWD at 14.64 g/kg/d (BHD-L group) from day 1 to day 52. Donepezil at the dose of 2.06mg/kg/d(donepezil group) was used as positive control. In steadily, mice in control group were injected intraperitoneally with saline and orally administered saline for 52 consecutive days.
